This can be achieved using 2 ways, 1st using poly line and 2nd using direction service provided by Google or other companies. In 2nd App, you want to know the distance between 2 moving vehicles. I am sure this can be achieved too using Google Services however I would like to know by distance, you mean road distance or distance between 2 points irrespective of road / track.
